CJC-1295
A synthetic growth-hormone-releasing hormone (GHRH) analogue with DAC (Drug Affinity Complex) modification for extended half-life and sustained GH release.
Mechanism of Action
CJC-1295 binds to GHRH receptors on anterior pituitary somatotrophs, stimulating growth hormone synthesis and release. The DAC modification (a maleimidopropionic acid linker) allows covalent binding to albumin, dramatically extending its half-life. This produces sustained, pulsatile GH elevation rather than an acute spike, more closely mimicking physiological GH secretion.
Key Published Studies
Prolonged Stimulation of Growth Hormone (GH) and Insulin-Like Growth Factor I Secretion by CJC-1295
Teichman SL, Neale A, Lawrence B, et al. — Journal of Clinical Endocrinology and Metabolism (2006)
A single injection of CJC-1295 DAC increased mean GH levels by 2-10 fold for 6 or more days and IGF-1 levels by 1.5-3 fold for 9-11 days with no serious adverse effects.
Effects of CJC-1295, a Long-Acting GHRH Analog, on GH Pulsatility and Sleep
Ionescu M, Frohman LA — Journal of Clinical Endocrinology and Metabolism (2006)
CJC-1295 increased GH pulsatile release and slow-wave sleep duration, suggesting beneficial effects on both somatotropic function and sleep architecture.
Dosage Range
CJC-1295 DAC: 1-2 mg once or twice weekly. CJC-1295 no DAC (mod GRF 1-29): 100-300 mcg 2-3 times daily.
Reconstitution
Reconstitute with bacteriostatic water. For a 2 mg vial, add 2 mL BAC water for a 1 mg/mL solution.
Storage
Store lyophilized at -20C. Reconstituted at 2-8C, use within 21 days.
Half-Life
CJC-1295 with DAC: ~6-8 days. Without DAC (mod GRF 1-29): ~30 minutes.
