NAD+ (Nicotinamide Adenine Dinucleotide)
A critical coenzyme found in every living cell, involved in energy metabolism, DNA repair, sirtuin activation, and aging research.
Mechanism of Action
NAD+ serves as a coenzyme for redox reactions in glycolysis, the TCA cycle, and oxidative phosphorylation. It is also a substrate for sirtuins (SIRT1-7), PARPs (poly-ADP-ribose polymerases), and CD38 — enzymes involved in DNA repair, gene silencing, calcium signaling, and circadian rhythm regulation. NAD+ levels decline with age, and restoring them is a major area of longevity research.

Nicotinamide Mononucleotide, a Key NAD+ Intermediate, Treats the Pathophysiology of Diet- and Age-Induced Diabetes in Mice
Yoshino J, Mills KF, Yoon MJ, Imai S — Cell Metabolism (2011)
NMN administration restored NAD+ levels in aged mice and improved glucose tolerance, hepatic insulin sensitivity, and lipid profiles.
Declining NAD+ Induces a Pseudohypoxic State Disrupting Nuclear-Mitochondrial Communication During Aging
Gomes AP, Price NL, Ling AJY, et al. — Cell (2013)
NAD+ decline during aging disrupts SIRT1-HIF1a signaling, contributing to mitochondrial dysfunction. Restoring NAD+ reversed these age-associated changes in mice within one week.
NAD+ Repletion Improves Mitochondrial and Stem Cell Function and Enhances Life Span in Mice
Zhang H, Ryu D, Wu Y, et al. — Science (2016)
NAD+ supplementation via NR improved mitochondrial function, muscle stem cell regeneration, and extended lifespan in aged mice.
Dosage Range
Research protocols typically use 250 mg to 1000 mg for in vitro and in vivo studies. Subcutaneous or IV administration in animal models.
Reconstitution
Dissolve lyophilized NAD+ powder in sterile bacteriostatic water. Highly soluble — a 1000 mg vial can be reconstituted in 5 mL for a 200 mg/mL solution.
Storage
Store lyophilized powder at -20C. Reconstituted solution at 2-8C, use within 14 days. Protect from light; NAD+ is photosensitive.
Half-Life
Circulating NAD+ has a half-life of approximately 30-45 minutes. Intracellular NAD+ turnover is tissue-dependent (1-10 hours).
