Tirzepatide
A dual GIP/GLP-1 receptor agonist studied for its effects on glycemic control and body weight reduction in metabolic research models.
Mechanism of Action
Tirzepatide activates both the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. This dual agonism enhances insulin secretion, suppresses glucagon release, slows gastric emptying, and acts on central appetite pathways in the hypothalamus to reduce food intake. The GIP component may also improve lipid metabolism and fat tissue signaling.
Key Published Studies
Tirzepatide Once Weekly for the Treatment of Obesity (SURMOUNT-1)
Jastreboff AM, Aronne LJ, Ahmad NN, et al. — New England Journal of Medicine (2022)
Participants receiving the highest dose of tirzepatide (15 mg) achieved a mean body weight reduction of 22.5% from baseline at 72 weeks compared to 2.4% with placebo.
Tirzepatide versus Semaglutide Once Weekly in Patients with Type 2 Diabetes (SURPASS-2)
Frias JP, Davies MJ, Rosenstock J, et al. — New England Journal of Medicine (2021)
Tirzepatide demonstrated superior HbA1c reduction and weight loss compared to semaglutide 1 mg across all dose levels in patients with type 2 diabetes.
Efficacy and Safety of Tirzepatide in Type 2 Diabetes (SURPASS-1)
Rosenstock J, Wysham C, Frias JP, et al. — New England Journal of Medicine (2021)
Tirzepatide monotherapy reduced HbA1c by up to 2.07% and body weight by up to 9.5 kg over 40 weeks compared to placebo.
Dosage Range
Research protocols typically range from 2.5 mg to 15 mg administered once weekly via subcutaneous injection.
Reconstitution
Reconstitute lyophilized powder with bacteriostatic water. For a 10 mg vial, add 1 mL BAC water for a 10 mg/mL concentration.
Storage
Store lyophilized powder at -20C to 4C. Reconstituted solution should be refrigerated at 2-8C and used within 28 days.
Half-Life
Approximately 5 days (120 hours), supporting once-weekly dosing protocols.
